(英文原文)anti-androgenic therapy with finasteride attenuates cardiachypertrophy and left ventricular dysfunction

methodsand results—we found 急性心力衰竭指南a strongly induced expressionof all three isoforms of the 5-α-reductase (srd5a1-3) in human and mouse heartswith pathological hypertrophy, which was associated with increased myocardialaccumulation of dht. starting one week after the induction of pressure overloadby transaortic constriction, mice were tre小儿心力衰竭的治疗ated with finasteride for two weeks.cardiac function, hypertrophy, dilation and fibrosis were markedly improved inresponse to finasteride treatment in male, but also in female mice. inaddition, finasteride also very effectively improved cardiac function andmortality after long-term pressure overload急性左侧心力衰竭 and prevented disease progressionin cardiomyopathic mice with myocardial gαq overexpression. mechanistically,finasteride by decreasing dht potently inhibited hypertrophy and akt dependent pro-hypertrophicsignaling in isolated cardiac myocytes, while introduction ofconstitutively active akt blunted 小儿心力衰竭these effects of finasteride.
